Tumor Immunity Assays
In Vitro (human and mouse cells):
1. Mixed Lymphocyte Reaction (MLR)
2. T-Cell proliferation
3. Maturation of antigen-presenting cells
4. Killing of target cancer cells by T-cells
In Vivo (mouse model):
Mouse melanoma model of lung metastasis. Immunocompetent mice are vaccinated with melanoma cell vaccine that partially stimulates anti-tumor immune response. This allows to observe and quantify co-stimulatory effects of anti-cancer compounds in all phases of anti-tumor immune response.
Example of MLR assay using lymphocytes labelled with green fluorescence dye CFSE to monitor proliferation of T-cells that were exposed to antigenic stimulus for 7 days. Cell proliferation, marker expression and viability were analyzed by flow cytometry.
