Advanced Models of Primary Tumors and Metastasis Initiated by Orthotopic/Ectopic Implantation and Treated with Potential Drugs

Metastatic progression is a series of low efficacy steps. It starts with the invasion of primary tumor into the vicinity of blood or lymphatic vessels, continues with cancer cells entering circulation and getting trapped in narrow capillaries in the target organ as dormant micrometastases. Finally, they eventually form actively growing macrometastases that kill patients. Anti-metastatic activity of a potential drug can attack any of these steps to bring clinical benefit. However, drugs targeting the actively growing metastasis are ultimately the most desirable ones. BioTest4U staff has experience with simple and complex analyses of metastatic progression and primary tumor growth. This service is devoted to our customers from Biotech/Pharma companies, but also Food Industry customers that are interested in testing the cancer prevention and intervention potential of their compounds and food supplements.

We also provide services that allow testing drug efficacy in primary tumor models and chemically-induced models for chemoprevention studies.

Metastatic Potential In Vitro

We provide analysis of metastatic potential in vitro by looking at single cell migration, collective cell migration, cancer cell proliferation and survival in the presence of normal host cells present in the organ where these metastasize. At the moment we provide a service called M-Spread Potential, which tests metastatic potential of cancer cells as well as anti-metastatic drug activity in 3D co-culture system with normal cells isolated from organs where breast cancer usually metastasize, i.e. lungs, liver, brain and bone.

Isolation and Quantification of Circulating Cancer Cells

We process and analyze mouse models and patient samples. We detect all types of carcinoma cells, cell clusters and cell ghosts (nuclei-free particles with cell membrane/cytoskeleton) that are present in the white-blood cell fraction. No negative depletion that may eliminate clusters of cancer cells attached to normal cells.

Chemosensitivity Testing

Personalized therapy is our ultimate goal.  We seek partnership with oncologists and other diagnostic laboratories to perform clinical trials and prospective studies on our chemosensitivity tests using live tumor samples. We provide testing in two- and three-dimensional cultures that mimic natural tumor growth, with or without normal cells that are likely to be part of the tumor in a given location, e.g. normal lung epithelial cells in the case of breast cancer that metastasize to lungs. We do small scale testing using conventional chemotherapeutic regimen and large scale testing using our chemical library.  Many of these drugs have been approved for indications other than cancer. Some have been shown to inhibit tumor growth in experimental animals or have a cancer preventive effect in humans.

Finally, we provide testing of selected candidate drugs in highly immunodeficient mice that are able to grow primary and metastatic samples from patients.
